摘要
目的 :探讨重组人胸腺素α原快速纯化方法 ,研究它对放射损伤免疫抑制小鼠免疫功能的调节作用。方法 :经超声碎菌、离心得上清、加热分离目的蛋白 ,用苯酚和氯仿抽提 ,最后经Super dex柱层析获胸腺素α原纯品。以 4.0Gy的剂量全身一次性照射致免疫功能低下小鼠为模型 ,观察腹腔注射胸腺素α原 30 0ng/ (只·d)对免疫功能的调节作用。结果 :胸腺素α原纯品可改善损伤小鼠的脱毛、体重减轻和尾部出血斑等一般状况 ,明显提高因损伤而萎缩的胸腺重量。虽不能影响外周血白细胞的总数 ,但可提高血淋巴细胞和血小板的数目及血红蛋白的量。结论 :纯化获得的胸腺素α原具有免疫调节活性 ,可促进放射损伤后免疫功能的恢复与重建。
Purpose:To establish an isolating and purifying method for recombinant prothymosin α, and to observe its immune regulating role in the irradiated mice.Methods:The recombinant prothymosin α was isolated and purified from bacterial proteins by heat denaturation,phenol and chloroform extraction and Super dex column 200 chromatography.The immune regulating role was experimented on immune suppressed mice induced by 4.0 Gy radiation.Results:The recombinant prothymosin α could be purified by the established method. It could improve the general status such as hair deprivation,body weight reduction and blood spots in the tails of irradiated mice. It could also increase the thymus weight after administering prothymosin intraperitoneally.It could not raise the number of white blood cells, but it could raise the number of lymphocytes and platelets and the content of hemoglobin.Conclusion:The recombinant prothymosin α had the immune regulating activity and it could promote the restoration and reconstitution of immune function in irradiated mice.
