摘要
目的观察穿山龙总皂苷(total saponin from Rhizoma Dioscorea nipponica,TSRDN)对Ⅱ型胶原诱导性关节炎(collagen-induced arthritis,CIA)大鼠血清中炎性细胞因子的影响,初步探讨穿山龙总皂苷治疗类风湿关节炎(rheumatoid arthritis,RA)可能的作用机制。方法建立CIA大鼠模型。于初次免疫后第14天,将造模成功的大鼠随机分为CIA模型组,穿山龙总皂苷高、中、低剂量组,雷公藤组。其中穿山龙总皂苷高、中、低剂量组分别经胃灌服100 mg.kg-1.d-1,50 mg.kg-1.d-1、25 mg.kg-1.d-1剂量的穿山龙总皂苷,雷公藤组灌服12 mg.kg-1.d-1剂量的雷公藤多苷,正常对照组、CIA模型组灌服等体积溶媒。用药后21 d,记录各组大鼠后肢体积、足垫厚度、关节炎指数;测定血清中TNF-α、IL-1β和IL-6水平;观察关节滑膜病理变化。结果经穿山龙总皂苷灌胃治疗21 d后,与模型组相比,治疗组大鼠后肢体积、足垫厚度、关节炎指数有降低趋势(P>0.05),大鼠滑膜增生减轻,炎细胞浸润减少,软骨及骨组织侵蚀破坏减轻,血清中TNF-α、IL-1β、IL-6水平明显降低(P<0.01)。结论穿山龙总皂苷能明显降低CIA大鼠血清中TNF-α、IL-1β和IL-6水平,减轻滑膜组织的病理损伤,表明其对治疗类风湿关节炎有一定作用。
Objective To study effects of total saponin from Rhizoma Dioscorea nipponica(TSRDN) on inflammatory cytokines in collagen-induced arthritis(CIA) rats,and investigate the possible mechanism of TSRDN in treating rheumatoid arthritis.Methods The CIA model was established.On the 14th day after the first immunization,the CIA rats were divided into 5 groups:the CIA model group,the three doses of TSRND groups(100,50,25 mg·kg-1·d-1) and the Tripterygium group(12 mg·kg-1·d-1).The rats of TSRND groups and Tripterygium group orally administrated with three doses of TSRDN and Tripterygium for 21 days.The rats of model group and control group were given the same volume of solvent.The arthritic index,volume and thickness of hinds were measured;the levels of TNF-α,IL-1β and IL-6 in the serum were detected by Enzyme-linked immunoadsordent assay(ELISA);the pathological changes of arthral synovial tissues were observed by HE staining with light microscopy.Results Compared with the model group,the arthritic index,volume and thickness of hinds appeared improving tendency in treated groups,but no statistical significance(P0.05);the synovium thickened slightly,the number of inflammatory cells decreased,the destruction of joint cartilage lighten;the levels of TNF-α,IL-1β and IL-6 in serum were significantly decreased(P0.01).Conclusion TSRDN could obviously decrease the levels of TNF-α,IL-1β and IL-6 in the serum,protected synovial tissues.The results suggested that TSRDN may be benificial for treatment of RA.
