镉损伤睾丸病理变化及蓝莓花青素保护作用 被引量:3

摘要 目的利用镉染毒小鼠模型研究蓝莓花青素(Ay)对镉染毒小鼠睾丸的保护作用。方法小鼠腹腔注射氯化镉和不同剂量花青素,14天后,观察镉致睾丸组织形态学改变,并通过测定NO含量及元素蓄积程度探讨镉致睾丸损伤的机制及Ay的保护作用。结果镉毒性能够引起损伤组睾丸曲精小管形态异常,生精细胞排列紊乱;而Ay能够减弱镉对睾丸的损伤作用。另外,镉能够在睾丸中蓄积,增加损伤组钙的含量,减少锌的水平,Ay能够减少镉在睾丸中的蓄积作用,缓解必需元素的改变;通过检测NO含量,发现镉的毒性作用可能与NO的过量产生有关,Ay能够减少NO的产生。结论镉通过在睾丸中蓄积,影响必需元素的浓度,抑制NO的产生,改变小鼠睾丸生精细胞的正常形态,损伤其正常机能;而Ay可能通过自由基清除、抑制NO以及金属螯合等作用起到保护作用。 Objective This assay is designed to study the effect of anthocyanins(Ay) on cadmium(Cd)-induced mice testis damage.Methods Mice were treated by i.p.given CdCl2 and different doses of Ay for 14 days,the histopathological changes can be observed using light microscope and the mechanism of Ay against Cd-intoxication was exploited by NO content determination and the accumulation of metal in vivo by atom absorbance spectrometry.Results The data showed that the intoxication of Cd led to the paramorphia of convoluted seminiferous tubule and the abnormal of spermatogenic cells,the effect of Ay can reduce these kinds of changes.Moreover,the accumulation of Cd in testis resulted in the increase of Calcium(Ca) concentration and the reduce of Zinc(Zn) levels,the administration of Ay can ameliorate the accumulation of Cd in testis and alter the concentration of Ca and Zn;the determination of NO content showed that the intoxication of Cd may relate to the overproduction of NO and the effect of Ay can decrease the NO content.Conclusion The absence of Cd in testis may cause the overproduction of NO,and alter the concentration of essential metals,as well as change the normal profile of testis,leading to the dysfunction of mice testis.However,the protection effect of Ay may be achieved by free radical scavenging,NO inhibition and metal chelating.
