摘要
目的:观察雄芍汤对肝星状细胞增殖和活化的影响。方法:实验设4组:空白组、空白血清组、扶正化瘀胶囊血清组、雄芍汤血清组。采用MTT比色法测定雄芍汤含药血清对体外培养的人肝星状细胞LX-2增殖的影响;免疫组织化学染色法检测LX-2中α-SMA、Ⅰ型胶原和Ⅲ型胶原的含量。结果:20%和10%扶正组和雄芍组对LX-2抑制率均高于相应浓度的空白血清组(P<0.01),5%扶正组、雄芍组和空白血清组差异不显著;雄芍各个浓度组对LX-2抑制率显著高于相应浓度的扶正组(P<0.05或P<0.01)。雄芍各组中,20%组和10%组对LX-2的抑制率差异不显著,但均显著高于5%组(P<0.01)。雄芍组和扶正组LX-2中α-SMA的含量显著低于空白血清组(P<0.01),扶正组α-SMA含量显著低于雄芍组(P<0.05)。雄芍血清组和扶正血清组LX-2中I型胶原、Ⅲ型胶原含量显著低于空白血清组(P<0.01),雄芍血清组和扶正血清组I型胶原差别不显著,扶正血清组Ⅲ型胶原含量显著低于雄芍血清组(P<0.05)。结论:雄芍汤能够抑制HSC的增殖和活化以及HSC中I型胶原和Ⅲ型胶原的分泌。
This study was aimed to observe effects of Xiongshao decoction on the multiplication and activation of hepatic stellate cell. This experiment contains 4 groups, which are blank group, blank serum group, Fuzhenghuayu capsule serum group (FZHY group), and Xiongshao decoction serum group (XS group). The determination was made on effects of Xiongshao decoction serum on multiplication of hominal hepatic stellate cells which are in vitro culture by MTT colorimetric method. And the α-SMA ,col- I and col-III contents were detected in LX-2 by immunohistostaining. The results showed that both LX-2 inhibition ratio of 20% and 10% FZHY group and XS group are higher than homologue concentration blank serum group (P 〈 0.01). And the difference among 5% FZHY group, XS group and blank serum group are not obvious. LX-2 inhibition ratio of each concentration XS group are higher than homologue concentration FZHY group (P 〈 0.05, or P 〈0.01). LX-2 inhibition ratio difference between 20% and 10% XS group are not obvious, but it is higher than 5% XS group (P 〈 0.01). The a- SMA content in LX-2 of FZHY group and XS group are lower than blank serum group (P 〈 0.01). And FZHY group is lower than XS group (P 〈 0.05). The col-I and col-III contents in LX-2 of XS group and FZHY group are lower than blank serum group (P 〈 0.01). The col-I difference between XS group and FZHY group are not obvious. And the col-III content in LX-2 of FZHY group is lower than XS group (P 〈 0.05). It was concluded that Xiongshao decoction can inhibit hepatic stellate cell to propagate and activate and secret col-I, col-III.
