摘要
目的探讨神经生长因子(NGF)对噪声所致听力损伤豚鼠的修复作用。方法取健康雄性豚鼠48只随机分成4组,分别为对照组(正常组)、腹腔注射组、穴位注射组、模型组(听力损伤),除对照组以外,其余3组豚鼠均24h放置在声压级100dB(A)的白噪声中,共7d,每组豚鼠均用畸变产物耳声发射(DPOAE)来判断其幅值、信噪比;然后通过注射NGF,观察其对各组豚鼠听觉通路上GJB2基因、SLC26A4基因表达的影响。结果穴位注射组DPOAE幅值测试结果和性噪比测试结果,提示除f0为0.25Hz处P>0.05外,其余频区P<0.01;穴位注射组GJB2基因、SLC26A4基因表达分别为8.45和6.65(OD/dL),显著低于其他观察组(P<0.05)。结论神经生长因子穴位注射不失为一种治疗噪声性听力损伤安全有效的新方法。
Objective To observe NGF on noise induced hearing impairment in guinea pigs repair. Methods Forty healthy male guinea pigs were randomly divided into 4 groups respectively,the control group (normal group),model group (hearing loss),acupoint injection group, the intraperitoneal injection group,except the control group,the remaining three groups of guinea pigs were exposed to 24 hours of sound pressure level 100 dB (A) white noise for 7 days. Each group of guinea pigs were used with distortion product otoacoustic emission (DPOAE) to determine its amplitude,signal to noise ratio. Results Acupoint injection in group DPOAE amplitude test results and the signal-to-noise ratio of the test results,suggesting that the remaining frequency zone P 〈 0.01 except f0 0.25 Hz (P 〉 0.05); GJB2 gene, SLC26A4 gene expression of point injection group were 8.45 and 6.65 (OD/dL), was significantly lower than that of other observa- tion groups, P 〈 0.05. Conclusion Nerve growth factor acupoint injection can be regarded as a safe and effective new treatment method of noise induced hearing impairment.
