低剂量硝酸镧对糖尿病大鼠肾脏结构的修复作用 被引量:7

摘要 目的通过观察低剂量硝酸镧对糖尿病大鼠肾脏形态结构的影响,探讨其对糖尿病大鼠肾脏结构损伤的修复作用。方法 Wistar雄性大鼠分为对照组、模型组(0.01 mol/L枸橼酸缓冲液诱导糖尿病模型)和硝酸镧组各10只。对照组和模型组每日给予生理盐水,硝酸镧组每日给予硝酸镧0.2 mg/kg,灌胃1个月。检测各组大鼠肾皮质厚度、肾小球体密度、肾小球总体积、近(远)端小管总体积、上皮细胞高度、管腔直径的变化及肾脏形态学改变。结果模型组大部分近端小管上皮细胞肿胀,远端小管部分扩张,硝酸镧组上述改变明显减轻。与对照组和硝酸镧组比较,模型组肾小球总体积及近(远)端小管总体积明显增大,近端小管上皮细胞高度明显增加,近(远)端小管管腔直径明显增大,差异均有统计学意义(P<0.01)。结论口服低剂量硝酸镧对糖尿病大鼠肾脏结构的损伤具有一定的修复作用。 Objective To explore the influence of low-dose lanthanum nitrate[La(NO3)3] on the morphological structure of the kidney of diabetic rats. Methods Male Wistar rats were randomly divided into control group, diabetic model group and La (NO3)3 treatment group, the rats in three groups were given daily saline, La (NO3)3 (0.2 mg/kg) through garage for one month. The kidney morphometry was used to detect the weight of each rat kidney, body weight, kidney weight and body weight ratio, renal cortical thickness, glomerular volume density, total glomerular volume, near (far) end of the total volume, near(far) end of the tubular epithelial cell height, near (far) end of tube cavity diameter changes and kidney morphological changes. Results Compared with control group, in diabetic model group, the kidney weight was increased significantly, the body weight was markedly decreased, the organ coefficients increased significantly, total glomerular volume, near (far) end of a small tube volume, proximal tubule epithelial cell height, near (far) end of the small tube cavity diameter were significantly increased all (P〈0.01); In La(NO3)3 treatment group, body weight reduced significantly(P〈0.01 ), the organ efficients and near (far) end of a small tube, total volume increased significantly (P〈0.01). HE staining results showed that the kidney in control and La (NO3)3 treatment groups were normal, the rat kidney in diabetic model group swelled; PAS staining results showed that most of the proximal tubular epithelial cells in diabetic model group swelled, containing PAS positive material increased; A little proximal tubules showed the shedding of epithelial cell, lumen expansion and part of expansion of the distal tubule; However no significant changes in epithelial ceils. Conclusion Oral administration of La(NO3)3 at low dose may present some recovery effect on the diabetic rat kidney structural damage.
