摘要
在常温下用吡嗪-2,3-二羧酸(C4N2H2(COOH)2,简称Pyzdc与金属镍反应制得新颖的吡嗪-2-3-二羧酸镍(Ⅱ)的配合物Ni[Pyzdc(COO)COOH]2.2H2O单晶.采用红外光谱、元素分析对化合物进行表征,应用X-射线单晶衍射技术测定配合物的晶体结构.配合物属于单斜晶系,P21/n空间群.2个吡嗪-2,3-二羧酸配体以氮原子和氧原子与镍(Ⅱ)离子反式螯合配位,2个H2O分子则以氧原子与镍(Ⅱ)离子轴向配位,形成八面体配合物.配合物分子之间通过氢键的弱相互作用构筑成一维、二维的空间结构.与此同时,应用紫外光谱法和荧光光谱法,初步研究配合物与核酸的作用方式,研究结果表明配合物与DNA主要以插入作用方式为主.进一步通过凝胶电泳法测定配合物对pBR322质粒DNA的断裂能力.
The novel complex Ni[Pyz (COO)COOH ]2·2HE2O was synthetized at normal temperature by the reaction of pyzdc and nickel chloride, where was pyzdc as 2,3-pyrazinedicarbaxylat and structurally characterized by IR spectra along with elemental analysis and X-ray single-crystal. The title complex belonged to monoclinic system with space group(P21/n). The center metal ion Ni(Ⅱ) rendered octahedron structure by N and O atoms from two Pyzdc molecules with trans-chelate way and O atoms from two H2O molecules with axial coordination. The complex formed 1D chain and 2D layer structure by hydrogen bonds interaction. Meanwhile the results showed that the complex bind to the DNA with intercalation mode which determined by UV spectrometry and fluorescent spectrometry. Furthermore capability of cleavage to pBR-322 plasmid DNA was confirmed by gel-electrophoresis assay.
