摘要
采用柱前衍生化高效液相色谱法,通过对PMP柱前衍生化温度、时间及色谱柱和流动相的考察,确定最佳检测条件,对10批样品的单糖组成进行分析,并运用中药指纹图谱相似度评价软件分析其结果。酵母多糖提取物总糖含量为84.20%,指纹图谱共检出3个峰,其中已通过对照品确定3个色谱峰的归属,分别为甘露糖、葡萄糖、半乳糖。10批酵母多糖水解物指纹图谱整体相似度良好,与共有模式之间相似度大于0.9。该方法简单、快速、灵敏度高、重现性良好,可作为控制酵母多糖提取物内在质量的标准。
Monosaccbaride Composition of the extract of zymosan was determined by HPLC after preparing precolumn derivitisation as PIMP derivatives. To determine the optimal testing conditions through time and temperature of derivitisation and the column and mobile phase selection. Results were analysed by the use of traditional Chinese medicine fingerprint similarity evaluation software. The total carbohydrate of the extract of zymosan was 84.20% .Fingerprint was composed of mannose, glucose, and galactose.The similarity of the 10 batch of samples to compare with common pattern was greater than 0.9. The method was a simple,rapid,sensitive and convenient, and can be applied to the quality control of the extract of zymosan. It could also be used to determine monosaccharide composition of acidic heteropolysaccbarides.
