参芪解郁方对产后抑郁大鼠行为学与脑组织形态学改变的干预研究 被引量:10

Research of influence of Shenqijieyu prescription on changes of behavioristics and morphology of brain tissue in rats with postpartum depression

摘要 目的探讨参芪解郁方对产后抑郁(PPD)模型大鼠行为学及脑组织形态学的干预作用。方法选用雌性2~3月龄SD大鼠,随机分为空白组、假手术组、模型组、中药组、西药组,每组再设1、2、4、6周观察时点,分别对大鼠进行行为学测试,并通过HE染色观察其脑组织神经细胞损伤情况。结果模型组各时点蔗糖水消耗较空白组降低(P<0.01,P<0.05),西药组第2周及中、西药组第4、6周较模型组升高(P<0.01)。模型组各时点敞箱测试(OFT)水平与垂直得分均较空白组降低(P<0.01,P<0.05),中、西药组除第1周外其他各时点水平得分与中药组第1周及中、西药组第2、4周垂直得分均较模型组升高(P<0.01,P<0.05)。模型组各时点强迫游泳测试(FST)不动时间较空白组延长(P<0.01,P<0.05),中、西药组各时点均较模型组缩短(P<0.01,P<0.05)。空白组前额皮质、海马、下丘脑细胞数量多、排列整齐、核仁清晰,模型组上述3个部位病理损伤第1、2周尤为显著,第4、6周亦无明显恢复。中、西药组病理损伤第1周时较模型组减轻,第2周时改善明显,第4、6周状况恢复基本接近正常水平。结论参芪解郁方对PPD模型大鼠抑郁行为有明显调节作用,同时可促进脑组织病理损伤恢复。 Objective To explore intervenfional effect of Shenqijieyu prescription on behavioristic and morphology of brain tissue in model rats with postpartum depression (PPD). Methods Female 2-3 month SD rats were selected and randomly divided into a blank group, a sham-operated group, a model group, a Chinese medicinal group and a western medicine group. The time points at 1st, 2nd, 4th, 6th week were set for observation in each group, at which behavioral test was applied respectively. The nerve cells injury in brain was also observed by HE stain. Results The sucrose consumption at each time point in the model group was decreased compared with that in the blank group (P 〈 0.01, P 〈 0.05); the consumption at 2nd week in the western medicine group and at 4th, 6th week in the Chinese medicinal group and the western medicine group was increased compared with that in the model group (P 〈 0.01). In the open field test (OFT), the scores of horizontal or vertical level at each time point in the model group were lower than those in the blank group (P 〈 0.01, P 〈 0.05); the scores of horizontal level at each time point (except the 1st week) in the Chinese medicinal group and the western medicine group, as well as the scores of vertical level at 1st week in the Chinese medicinal group and at 2nd, 4th week in the Chinese medicinal group and the western medicine group were higher than those in the model group (P 〈 0.01, P 〈 0.05).The immobility time in forced swimming test (FST) at each time point, in the model group were longer than that in the blank group (P 〈 0.01, P 〈 0.05); in the Chinese medicinal group and the western medicine group were shorter than that in the model group (P 〈 0.01, P 〈 0.05). The cells of prefrontal cortex, hippocampus and hypothalamus in the blank group were plenty and arranged with clear nucleolus, the pathological injury of above three, locations in the model group was particularly apparent at 1st, 2nd week and there was not obvious recovery at 4th, 6th week. The pathological injury in the Chinese medicinal group and the western medicine group at 1st week was relieved compared with that in the model group, which was improved apparently at 2nd week and recovered almost to the normal level at 4th, 6th week. Conclusion The Shenqijieyu prescription has an obvious regulating action for the depressive behavior in PPD model rats, which could improve the recovery of pathological injury in brain tissue at the same time.
