摘要
以益母草根颈为材料,进行了愈伤组织诱导、增殖与分化,分化芽生根,试管苗移栽、定植的研究.结果表明:MS+6-BA 0.5mg·L-1+NAA 0.6mg·L-1+2,4-D1.2mg·L-1是愈伤组织诱导培养的理想培养基;MS+6-BA0.5mg·L-1+NAA0.1mg·L-1+2,4-D 1.5mg·L-1是愈伤组织继代增殖培养的理想培养基;1/2MS+6-BA 0.2mg·L-1+NAA 0.1mg·L-1是愈伤组织分化培养和分化芽继代分化增殖培养的理想培养基;N6+IAA0.2mg·L-1培养基是益母草分化芽生根培养的理想培养基.移栽成活率为94.4%;定植成活率为99.1%;定植成活的试管苗保持野生益母草的所有植物学性状.
In order to preserve the resources and meet the needs of planting,the rhizomes of Leonurus japonicus were used as explants to do the research on callus inducement, multiplication and differen- tiation,rooting of differentiation buds, ransplantation of tube seedlings and stable planting. The re- sults showed that the optimum medium for callus induction was MS+6-BA 0.5mg·L-1+NAA 0.6mg·L-1+2,4-D1.2mg·L-1;MS+6-BA0.5mg·L-1+NAA0.1mg·L-1+2,4-D 1.5mg·L-1 was the ideal medium for callus multiplication; 1/2MS+6-BA 0.2mg·L-1+NAA 0.1mg·L-1 was suitable for callus differentiation and multiplication of differentiation buds; the best medium for roo- ting of differentiation buds was N6+IAA0.2mg·L-1. The transplanting survival rate was 94.4% and stable planting survival rate was 99.1% ; the tube seedlings stable planted maintained all botany characteristics of the wild Leonurus japonicus.
