摘要
用薄层色谱法分析烷基糖苷APG-06,选出了最佳展开剂,并用高效液相色谱进行验证,对分离的组分进行核磁表征和分析,计算样品聚合度(n)。结果表明,以氯仿/无水乙醇/氨水=9.5∶6.5∶0.5(v/v/v)作为展开剂的薄层色谱的分析结果与HPLC一致。分离的APG-06经核磁(1H NMR)计算得到聚合度为1.00,二苷~多苷的平均聚合度为3.47。薄层色谱的优点在于分离能力强,仪器简单,廉价,操作方便。
The short chain of alkyl polyglucosides (APG-06) was separated and analyzed by the thin layer chromatography (TLC). The optimum developer was chosen by the HPLC on a verification, 1H NMR characterization and analysis of the separated components, the sample was calculated degree of polymeri- zation. The results showed that mobile phase of CHC1/CH3CH2OH/NH30H (9.5: 6.5:0.5 ,v/v/v), and the result of TLC identified with HPLC. The alkyl monoglucoside can be separated, and the degree of polymerization is 1.00, and the alkyl polyglucosides is 3.47. The feature of good separating, easier operating, lower-cost make the TLC be convenient for chemical plants to purify the industrial grade APG.
