摘要 探索一种简便、灵敏度高、特异性强且易推广的血清中突变型p53蛋白检测方法 ,用于肿瘤早期诊断。用突变型p53蛋白的单克隆抗体 ,与特定DNA片段连接制成基因探针 ,用免疫 -PCR技术 ,对 117例病人血清样品进行检测。在 117例病人血清样品中 ,检测总阳性率为 4 1 88% ( 4 9/ 117) ,在临床怀疑为癌症及肿瘤病人中阳性率达 52 11% ( 37/ 71)。 To investigate immuno-polymerase chain reaction system,a simple,sensitive,specitic detection of mutant p53 protein for early prognosis in tumor.The probe is made by specific antibody-DNA conjugates.Using the immuno-PCR system to detect mutant p53 protein in human serum.Results in 117 serum samples,the total positive rate was 41.88%(49/117).The positive rate in the samples which were suspected as cancer was 52.11%(37/71).The research suggested the immuno-PCR system is simple,sensitive,specific for early prognosis in tumor and practicable for general survey of such disease.Also it can be used for more genic diseases.
