摘要
目的:评价两种可降解型种植体支抗材料聚乳酸酯和羟基磷灰石对人成骨样细胞MG-63的细胞毒性,检测OPG/RANKL mRNA的表达量。方法:选择人成骨样细胞MG-63作为细胞毒性的实验对象。MTT法检测细胞活力,评价两种可降解型种植体支抗材料聚乳酸酯和羟基磷灰石微粒对人成骨样细胞增殖的影响。用realtime-PCR检测人成骨样细胞内OPG/RANKL mRNA的表达量。结果:钛粒子对细胞生长具有一定的抑制作用,羟基磷灰石和聚乳酸酯颗粒对细胞的增殖没有明显的副作用。钛粒子可以有效的诱导成骨样细胞OPG的表达,同时也增强了RANKL的表达,并随着粒子作用时间的增加,作用效果也相应增加。与钛粒子相比较,羟基磷灰石和聚乳酸酯颗粒对细胞OPG/RANKL mRNA表达无明显的影响,且随着作用时间的增加及在不同浓度的干预下,细胞OPG/RANKL mRNA表达也无明显的影响。结论:聚乳酸酯和羟基磷灰石对破骨细胞增殖和OPG/RANKL mRNA的表达无明显影响。
Objective: To evaluate the cytotoxicity of BIOS mater/al and exam the expression of OPG/RANKL mRNA in human osteoblast-like cells MG-63 with polylaetic acid and hydroxyapatite microparticle. Methods: Human osteoblast-like cells MG-63 was chosen to evaluate the cytotoxicity of polylactic acid and hydroxyapatite. The proliferation of osteoblast was detected by MIT method. The expression of OPG/RANKL mRNA was examined by realtime-PCR. Results: MIT test demonstrated that titanium particles can obviously degrade the cell viability of osteoblast, while PLA and HAP showed no inhibited effect. Titanium particles can increase OPG/RANKL mRNA expression. Compared to the titanium parti- cles, PLA and HAP have no effects on the expression of OPG/RANKL mRNA. Conclusion: PLA and HAP microparticle has no effects on the human osteoblast-like cells proliferation and OPG/RANKL mRNA expression.
