摘要
目的筛选大鼠痛风性关节炎(GA)生物标志物。方法采用改良的微晶尿酸钠和次黄嘌呤法制备大鼠急性GA模型,检测血尿酸、尿尿酸、24 h总尿量及滑膜组织病理形态学改变,采用RayBioBiotin Label-based Rat Antibody ArrayⅠ筛选差异表达蛋白。结果急性GA模型建立成功,发现GA中14个差异表达蛋白,其中9个细胞因子首见于GA,差异最明显2个蛋白为TRAIL与Neuropilin-2。结论 TRAIL与Neuropilin-2为GA生物标志物,为临床诊断与防治提供新靶点。
Aim To determine specially expressed pro-teins by testing proteins of synovium in AGA rats.Meth-ods The animal model of AGA was set up by Monosodium urate crystal(MSU) combined with hypoxanthine(HX),which was modified in our previous experiment.Blood and Urine samples were used to measure serum uric acid,urine uric acid and 24 h total volum of urine,and synovium tissues were assessed by histological staining and protein quantitation.Three chips of RayBio Human Label-based Antibody Array Ⅰ were applied to detect 90 proteins in synovium tissue of AGA rats.A total of 14 differentially expressed proteins werefound in the synovium of AGA rats,and nine of them were first found in this model.There were seven upregulated and seven down-regulated proteins.TRAIL and Neuropilin-2 are most differentially expressed proteins.Conclusions Both TRAIL and Neuropilin-2 can be identified as key contributors to the pathomechanism of AGA,and they are biomarkers of gouty arthritis in clinical diagnosis and treatment of the disease.
