摘要
目的将一种硬性接触镜护理液(RCLS)与现有同类产品Menicon Progent进行除蛋白作用、对人眼角膜上皮细胞毒性及动物刺激性的比较,综合评价其生物相容性。方法细胞毒性实验采用人眼角膜上皮细胞,分别用培养液、RCLS、Menicon Progent作用细胞不同时间,以WST-1试剂检测细胞存活率;取硬镜RGP镜片在2mg/mL溶菌酶溶液中浸泡、吸附24h,分别用RCLS和Menicon Progent浸泡吸附后的镜片,再用BCA法测定溶液中的蛋白含量;刺激性实验以新西兰白兔为实验对象,分别取0.1mLRCLS或Menicon Progent滴入兔眼,记录眼损伤;取沾有RCLS和Menicon Progent的纱布块敷贴于兔背部裸露皮肤处,敷贴后评价皮肤损伤程度。结果 RCLS对硬镜表面吸附的溶菌酶有83.159%的除去率,相对于Menicon Progent的85.180%,差异无统计学意义;Menicon Progent接触15min后即对人角膜上皮细胞有溶解作用,细胞毒性大,对兔眼有刺激,对皮肤也有轻度刺激,而RCLS作用人角膜上皮细胞1h,细胞存活率>80%,对兔眼及皮肤均无刺激。结论 RCLS在细胞毒性及刺激性方面的生物相容性优于Menicon Progent。
Objective To evaluate the biocompatibility of a new type of rigid contact lens care solution(RCLS) including deproteinization effect, cytotoxicity on human corneal epithelium cells, irritation of eyes and skin, compared with MeniconProgent, a commercially available contact lens care solution. Methods The human corneal epithelial cells HCE-2 were planted in 96-well plates, co-cultured with RCLS, MeniconProgent and cell medium respectively. Cell viability was determined using WST-1 reagent to detect the absorbance of each well after exposure for different time at 450 nm; For deproteinization efficacy experiment, two groups of rigid contact lens were soaked in 2 mg/mL lysozyme solution for 24 hours, then immersed in RCLS or MeniconProgent for 30 min. The protein content of each solution was determined with BCA kit; The New Zealand white rabbits were chosen to take irritant experiment. Each rabbit had one drop testing solution in one eye. The result was recorded after experiment. The gauze pieces was steeped with RCLS or MeniconProgent before fixing them onto the skin of rabbits' back, and the injury of the skin was evaluated. Results The RCLS had a 83. 159% deproteinization rate of lysozyme adsorbed upon the surface of lens, comparing to MeniconProgent of 85. 180%, which had no significant difference. Cytotoxicity results showed that MeniconProgent might lead to cell lysis which reveals a severe toxicity on corneal epithelium cells,and it also has irritating effect to eyes and skin. When co-cultured in RCLS for an hour, the toxicity of RCLS was less than 20%, neither irritating to the rabbit eyes nor to the skin. Conclusions RCLS had a superior biocompatibility comparing to MeniconProgent in regard to cytotoxicity and irratanting. ( Chin J Ophthalmol and Otorhinolaryngo1,2013,13 : 100-104)
