摘要 背景与目的:研究水通道蛋白-1(AQP-1)与血管内皮生长因子(VEGF)在血管母细胞瘤中的表达,并探讨其在血管母细胞瘤生成中的作用。方法:应用免疫组化方法,并通过测量积分光密度值(IOD)研究AQP-1与VEGF在30例囊性血管母细胞瘤、10例实体性血管母细胞瘤以及10例正常小脑组织中表达情况。结果:AQP-1与VEGF在30例囊性以及10例实体性血管母细胞瘤中表达显著增强(P<0.05),且AQP-1在囊性肿瘤中的表达高于实体性肿瘤。结论:AQP-1的高表达可能在维持肿瘤细胞的高代谢需求中起促进和维持肿瘤增殖的重要作用;VEGF则在肿瘤血管生成中起重要作用,而且与AQP-1协同作用,共同促进肿瘤的增殖和血管的生成。 BACKGROUND & OBJECTIVE: The mechanisms of development of hemangioblastoma is not fully understood. In this article, we observed the expression level of aquaporin-1 (AQP-1) and vascular endothelial growth factor (VEGF) in hemangioblastomas, and investigated the effects of AQP-1 and VEGF to the formation of hemangioblastomas. METHODS: The protein expression level of AQP-1 and VEGF were examined by immunohistochemical method in 30 cystic hemangioblastomas and 10 solid hemangioblastomas and 10 normal cerebellum tissue. RESULTS: The expression level of AQP-1 and VEGF increased obviously in hemangioblastomas compared with that in normal brain tissue, and the expression level of AQP-1 was more in cystic hemangioblastomas than that in solid hemangioblastomas. CONCLUSIONS: Significant up-regulation of AQP-1 expression may play an important role in maintaining the high metabolic turnover and proliferation in hemangioblastomas. The high expression level of VEGF may play a role in the progress of angiogenesis and cooperates with AQP-1 to enhance the tumor's proliferation and angiogenesis. The expression of AQP-1 in normal cerebellum may help to explain the reason about the predilection site of hemangioblastomas is cerebellum.
