摘要 为了检测大麦提取物对自由基DMPD.+的清除作用以及对蛋白质、DNA和细胞DNA氧化损伤的保护作用,将大麦提取物加入DMPD反应体系、AAPH引发的蛋白质和DNA氧化损伤体系、H2O2引发的细胞DNA氧化损伤体系中,分别以比色法检测产物生成、SDS-PAGE和琼脂糖凝胶电泳、彗星电泳法检测损伤程度.结果表明,大麦提取物可有效清除DMPD.+自由基,125μg.mL-1时清除率超过50.0%,250μg.mL-1时对AAPH引起的蛋白质氧化损伤的保护率约为67.0%,125μg.mL-1时几乎完全保护了DNA不受损伤;在保护细胞DNA时,其作用与Trolox相同,1 000μg.mL-1时保护率可达87.9%.说明大麦提取物在各个氧化体系中都表现出突出的抗氧化活性,是优秀的天然抗氧化剂. To evaluate the scavenging effect on free radical DMPD"+, the protection effect on protein, DNA and cell DNA oxidative damage of barely acetone extract, different concentration barely extract are added to DMPD system, AAPH-induced protein and DNA damage system, and H202-initiated cell DNA damage system, respectively. The scavenging effect of barely extract on free radical is examined by the colorimetric determination, the protection effect on oxidative damage of DNA, protein and cell DNA damage are tested by SDS-PAGE, agarose gel electrophoresis and comet assay. The results show that barely extract scavenge DMPD+ effectively, the scavenging rate exceed 50.0% at 125 μg · mL^-1, the protection rate to AAPH-induced protein oxidative damage is 67.0% at 250μg · mL^-1, and almost protect DNA completely at 125 μg · mL^-1 Its activity is as same as Trolox's when protecting cell DNA, the protection rate reached 87. 9% at 1 000 μg · mL^-1. These results indicate that barely extract possess remarkable antioxidative activities in these systems and is an excellent nature antioxidant.
