摘要
目的:获得ε-聚赖氨酸产生菌的最佳发酵培养基。方法:以白色链霉菌N31-69菌株为研究对象,以各种碳源、氮源、无机盐为主要考察因素,分析各因素对ε-聚赖氨酸合成的影响,确定最佳碳源和最佳氮源。在单因素试验的基础上进行正交试验,确定最佳发酵培养基。结果:最佳发酵培养基组成份(%):葡萄糖3,(NH4)2SO40.7,K2HPO40.08,KH2PO40.17,MgSO4.7H2O 0.07,ZnSO4.7H2O 0.012,FeSO4.7H2O 0.003,酵母浸出粉0.7。N31-69菌株在最佳发酵培养基中,ε-聚赖氨酸摇瓶发酵产量达1.519 g/L,比优化前产量提高了28.0%。结论:今后在发酵罐上进一步进行发酵优化提高产量,为ε-聚赖氨酸产业化打下基础。
Objective:To obtain the optimum fermentation medium of ε-polylysine.Methods:The effects of carbon sources、nitrogen sources、mineral salts on ε-polylysine biosynthesis in shake flasks were studied to obtain the best sources of carbon and nitrogen.The optimum medium was found by the orthogonal test.Results:Through the orthogonal test,we found the optimum medium was 3% glucose,0.7%(NH4)2SO4,0.08% K2HPO4,0.17% KH2PO4,0.07% MgSO4·7H2O,0.012% ZnSO4·7H2O,0.003% FeSO4·7H2O,0.7% extract-yeast.Under the optimum medium,the productivity of ε-polylysine by strain N31-69 could reach 1.519 g/L,increased by 28.0% compared with the original medium.Conclusion:The fermentation medium in bioreactor to increase the productivity of ε-polylysine can be optimized,which would pave a way for the industrialization of ε-polylysine.
