IL-22基因单核苷酸多态性与肝癌易感性的相关性研究 被引量:5

摘要 目的:利用高通量飞行时间质谱来探讨IL-22基因多态性与肝癌易感性的相关性。方法:选取2011年1月至2012年12月赣南医学院第一附属医院普外科收治的病理确诊的肝癌患者55例,同期在本医院体检科体检的健康人60例为对照组,采用美国Sequenom公司建立的Massarray检测平台分析IL-22基因4个SNP位点(rs2227472、rs2227478、rs2227483、rs2227473)基因型,统计两组人群各个SNP等位基因频率;应用ELISA方法检测不同基因型肝癌患者外周血单个核细胞(PBMC)的培养上清液中IL-22浓度。结果:IL-22基因rs2227472位点存在GG、GA和AA 3种基因型,病例组等位基因G频率(57.2%)高于健康对照组(51.7%),差异有统计学意义(P<0.05),其他3个位点病例组和对照组之间差异无统计学意义;rs2227472位点GG基因型肝癌患者的PBMC培养上清液中IL-22浓度为(40±6)ng/L,明显高于AA/GA基因型肝癌患者(17±5)ng/L,差异有统计学意义(P<0.05)。结论:IL-22基因rs2227472多态性与肝癌易感性有相关性,其等位基因G可能和肝癌发生有关;rs2227472多态性影响机体PBMC的IL-22的分泌,在肝癌的免疫学机制中起关键作用。 Objective:To study the correlation between the SNP of interleukin-22 and susceptibility to HCC with High throughput MALDI-TOF.Methods:From January,2011 to December,2012,55 patients with HCC in general surgery department and 60 healthy doners in physical examnation department of The First Affiliated Hospital of Gannan Medical University were selected.The polymorphism of interleukin-22 gene(rs2227472、rs2227478、rs2227483、rs2227473) was detected by using USA Sequenom Massarray,SNP allelic frequencies between two groups were statisticed by chisquare;IL-22 concentration of different genotypes in PBMC culture supernatant was detected by ELISA,and analyzed by t-test.Results:The genotypic frequency of G of IL-22(rs2227472) in the patient groups(57.2%) were significantly higher than that of the control groups(51.7%)(P<0.05).IL-22 concentration of genotypes GG.HCC patients PBMC culture supernatant(40±6) ng/L was statistically higher than that of AA/AG groups(17±5)ng/L(P<0.05),No significant difference was found in other 3 SNPs of IL-22 between patient groups and control groups.Conclusion: The polymorphism of IL-22(rs2227472) gene may be associated with susceptibility to HCC.G allele gene may be related to the occurrence of HCC.rs2227472 polymorphisms affected IL-22 secretion of the body PBMCs,playing a key role in HCC immunological mechanisms.
