高效液相色谱法检测牙周炎患者基因组整体甲基化水平的研究

摘要 目的比较慢性牙周炎人群与牙周健康人群基因组整体甲基化水平的差异,探讨基因组整体甲基化水平与慢性牙周炎易感性的相关性。方法慢性牙周炎患者46例作为牙周炎组,牙周健康者37人作为对照组。颊黏膜细胞拭子法提取样本DNA,高效液相色谱法检测基因组5-甲基脱氧胞苷酸含量。结果牙周炎组5-甲基脱氧胞苷酸含量为(3.360±0.023)%,对照组为(3.428±0.023)%,组间差异没有统计学意义(P>0.05)。结论尚不能认为基因组整体甲基化水平改变是慢性牙周炎的危险因素。 Objective Analyse the differences of the overall methylation level of genome among patients with peri- odontitis and healthy people, and find out the relation between genomic overall methylation level and susceptibility of pe- riodontitis. Methods Periodontitis group were 46 patients with periodontitis and healthy group were 37 cases of healthy people for control. DNA was taken by swabbing buecal mucosa exfoliated cells with cotton and was detected by high per- formance liquid chromatography ( HPLC ), Results The content of 5- methyl deoxycytidine in periodontitis group ge- nome was (3. 360± 0. 023 ) % and healthy group was ( 3. 428± 0. 023 ) %. There was no statistically significance be- tween two groups ( P 〉 0.05 ). Conclusion The overall genome methylation level does not indicate that changes in the overall methylation level detected by HPLC are risk factors for periodontitis.
