毛细管电泳法测定去甲肾上腺素进样方法探讨

摘要 目的 :比较毛细管电泳法测定去甲肾上腺素 (NE)中流体力学和区带浓缩两种进样方法对检测灵敏度的影响。方法 :区带浓缩法 :2mmol·L-1硼酸压力进样 10s,样品电进样 80s ,(样品溶于 2mmol·L-1硼酸溶液中 ) ;流体力学法 :样品压力进样 80s(样品溶于 2 0mmol·L-1硼酸溶液中 )。结果 :区带浓缩法和流体力学法的检测限分别为NE1ng·ml-1与 1μg·ml-1。结论 :区带浓缩法进样有很高的浓缩因子 ,检测限从流体力学法的 1μg·ml-1降至 1ng·ml-1,灵敏度提高了 10 0 0倍。 Aim To compare the impact of stacking and flow injection on capillary electrophoresis for the determination of noradrenaline (NE).Methods Stacking injection:2mmol·L -1 Boric acid was injected by pressure for 10sec, sample was injected by voltage for 80 sec(sample solute in 2mmol·L -1 Borid acid); hydrostatic injection: Sample was injected by pressure for 80 sec(sample solute in 20mmol·L -1 Boric acid).Results The detection limits of stacking and hydrostatic injection of NE were 1ng·ml -1 and 1μg·ml -1 respectively.Conclusion Stacking injection has high condensing factor,its sensitivity was 1000-fold higher than hydrostatic injection.
