黄芩甙对小鼠肝细胞色素P_(450)及其亚家族的诱导

Effects of baicalin on liver microsomal cytochrome P_(450) system

目的 :观察黄芩甙对小鼠肝细胞色素P4 50 及其亚家族的影响 ,深入探讨黄芩甙保肝作用的机理。方法 :采用分光光度法分别测定小鼠肝微粒体细胞色素P4 50 、b5含量及氨基比林N -脱甲基酶 (ADM)、7-乙氧基香豆素O -脱乙基酶 (ECD)、苯并芘羟化酶 (AHH)。采用蛋白印迹杂交技术鉴定细胞色素P4 50 同功酶。结果 :给小鼠黄芩甙 (10 0mg·kg-1·d-1)灌胃 ,连续 7d ,可使小鼠肝微粒体细胞色素P4 50 含量显著增加 ,并使ADM、ECD及AHH 3种酶活力显著增强。对 6种同功酶的鉴定结果显示 ,黄芩甙可诱导 1A1、2B1及 2C113种同功酶 ,对细胞色素b5含量及 3A2、2D1、2E13种同功酶无诱导作用。结论 :黄芩甙可显著诱导小鼠肝细胞色素P4 50 的含量和活性 ,对其亚家族的诱导以 1A1、2B1及 2C11为主。

Aim To investigate the effects of Baicalin on liver microsomal cytochrome P 450 system.Methods Liver microsomal cytochrome P 450 ,b 5, aminopyrin N-demethylase(ADM), 7-ethoxycoumarin O-deethylase(ECD), and Benzopyrene hydroxylase (AHH) activity were determined by UV chromatography.Activities of six cytochrome P 450 isoforms were assayed with Western Blotting.Results The liver microsomal cytochrome P 450 level and ADM, ECD and AHH activity were increased significantly by baicalin treatment. The three P 450 isoforms, 1A1, 2B1 and 2C11, were also selectively induced by Baicalin.Conclusion Baicalin increases liver microsomal cytochrome P 450 level and selectively induces 1A1, 2B1 and 2C11 of P 450 isoforms in mice.
