摘要
目的分析CD4+CD25+Foxp3+调节性T淋巴细胞(Treg)数量和分布的相关性,研究两者之间的相关性。方法选取该院60例胃癌患者,用流式细胞仪分析患者术前外周血与肿瘤浸润淋巴细胞(TIL)中Treg以及CD8+T细胞的比率。另选出10例胃癌患者分别于手术前、后用ELISA检测血清和腹腔冲洗液中细胞因子的水平,并进行对比分析。结果胃癌患者外周血分析CD4+CD25+Foxp3+Treg显著高于健康对照组,癌症早期患者低于晚期患者(P<0.05);胃癌患者血清比例低于健康对照组(P<0.05)。肿瘤组织浸润淋巴细胞中CD4+CD25+Foxp3+Treg的比率与CD8+T细胞的比率呈负相关(r=-0.4,P<0.05)。10例患者肿瘤切除后CD4+CD25+Foxp3+Treg的比率显著降低(P<0.01)。结论胃癌患者外周血中的CD4+CD25+Foxp3+Treg明显比CD8+T细胞高很多,发展速度极快,经分析研究,CD4+CD25+Foxp3+Treg比率的增高与CD8+T细胞的降低,可能与癌细胞自身的免疫力有着密切关系。
Objective To analyze CD4+CD25+Foxp3+regulatory T(Treg)lymphocyte numbers and distribution,to study the relationship between the two.Methods 60patients with gastric cancer in the hospital were enrolled in the study,the multi-parameter flow cytometry analysis were applied for the Treg and CD8+ T cell ratio in the peripheral blood and tumor infiltrating lymphocytes(TIL)for preoperative patients.10patients with gastric cancer were recruited and had ELISA detection of IFN-γ,IL-4cytokines extent and comparative analysis before and after operation.Results The patients with gastric analysis of CD4+ CD25+ Foxp3+regulatory T lymphocytes was significantly higher than the healthy control group,lower than that of patients with advanced disease(P0.05).For patients with early cancer,proportion of patients with gastric cancer serum Th1/Th2was lower than the healthy control group,the difference was statistically significant(P0.05).Tumor infiltrating lymphocytes CD4+ CD25+Foxp3+ regulatory T lymphocyte ratio and CD8+ T cell ratio was negatively correlated(r=-0.4,P0.05).10patients after removal of tumor,CD4+ CD25+Foxp3+regulatory T lymphocytes ratio was significantly reduced,there were differences with statistically significant(P0.01).Conclusion patients with gastric cancer in peripheral blood CD4+ CD25+Foxp3+regulatory T lymphocytes was significantly higher than that CD8+T cells,and the fast progress analysis study,CD4+CD25+Foxp3+regulatory T lymphocyte ratio increased and CD8+ T cells reduced,probably has a close relationship with the cancer cell's own immune system.
