Evaluation of glomerulosclerosis progression in the PAN rat model by measuring the glomerular area

摘要 目的研究不同时间点嘌呤霉素氨基核苷(PAN)大鼠肾病模型的肾小球面积,及其与肾小球硬化指数之间的关系,评估肾小球硬化的进展过程。方法成年雄性SD大鼠,体重200~250 g。PAN组大鼠尾静脉注射PAN,对照组注射等量生理盐水。成模后于4、14及20周处死大鼠,取其肾脏,石蜡包埋,切片,高倍镜下观察拍片,测定肾小球面积及硬化指数,进行统计学分析并观察肾脏病理组织变化。结果 PAN组在4周时肾小球面积与对照组比较差异无统计学意义(P>0.05);14周时,PAN组明显>正常对照组(P<0.01);但是在20周时PAN组肾小球面积<对照组(P<0.001),且20周时肾小球硬化指数与肾小球面积之间的相关系数为-0.82,呈强负相关关系。结论肾小球面积测定在评估PAN诱导肾炎肾小球硬化的发展中起重要作用。疾病的早期小球面积代偿性增大,随着疾病的进展、肾小球的面积随着硬化程度的增加而减小,与肾小球硬化指数存在强负相关关系。因此,观察肾小球面积的改变可以反映肾小球疾病进展及肾小球硬化的典型过程。 Objective To evaluate the glomerulosclerosis progression in the puromycin aminonucleoside (PAN) nephropathy by measuring glomerular area (GA). Methods PAN model was made by injection of PAN via tail vein to male aduh SD rats; and rats in controls were injected with 0.9% NS. The rats were executed at 4 weeks, 14 weeks and 20 weeks and kidneys were taken out and embedded in paraffin for making the slices. The glomerular areas were measured in high magnification observation, and analyzed the relationship with glomerulosclerosis index (GI) by using binary variable correlation analysis. Results There was no significant changes at 4 weeks comparing with rats in controls in average glomerular area in PAN rats; the average GA in the PAN rats was signifi- cantly bigger than that in the control group at 14 weeks( P 〈 0.01 ) ; but the average GA in the PAN rats was significantly smaller than that in the control group at 20 weeks ( P 〈 0. 001 ) , there was the negative correlation between GA and GI at 20 weeks ( r = - O. 82 ).Conclusion The measurement of GA plays an important role in the evaluation of the glomerulosclerosis in the PAN nephropathy devel- opment. There is a strong negative correlation between GA and GI. Therefore, the size of glomerular area can indicates the typical pro- gression process of glomerularsclerosis in the renal disease.
