摘要
取厚叶岩白菜优良单株茎尖作为外植体,试验结果表明:在改良MS培养基上,当NAA浓度为0.1 mg/L,BA浓度取1.0 mg/L,添加PVP 0.5 mg/L时,月增殖系数为3.03;在培养基1/2MS+IBA1.0 mg/L+PVP 0.5 mg/L中生根效果明显,生根率达90%;在腐殖土、黄沙、珍珠岩体积比为2∶1∶1的混合基质中,移栽成活率达90%。建立了高效的厚叶岩白菜离体培养快繁技术体系。
Tissue culture of Bergenia crassifolia ( L. ) Fritsch was developed by organogenesis with apical buds as the explants. The results were that, the efficient multiplication medium was reformed MS supplement with O. 1 mg/L NAA, 1.0 mg/L BA, and 0.5 mg/L PVP, the average multiplication rate was 3.03. 1/2MS with IBA 1.0 mg/L and PVP 0.5 mg/L was the optimal rooting medium, and the rooting rate was about 900. Acclimatized the regenerated plants in medium of muck, yellow sand and perlite (2: 1:1 ) , the survival rate reached to 90%. Therefore, the rapid propagation system for Bergenia crassifolia was established.
