摘要
目的研究IRE-BP 1蛋白与Bel-7402肝癌细胞耐药性的关系。方法根据已知IRE-BP 1蛋白序列设计引物扩增基因片段,荧光定量PCR检测其在敏感Bel-7402细胞株、阿霉素抗性Bel-7402细胞株之间的表达水平差异。结果成功扩增Bel-7402肝癌细胞IRE-BP 1蛋白基因片段,荧光定量PCR表明,IRE-BP 1蛋白基因在阿霉素抗性Bel-7402细胞株的表达量是敏感Bel-7402细胞株的3.57倍。结论 IRE-BP 1蛋白基因在肝癌细胞耐药发生发展机制中存在一定的作用,可为新的耐药性治理备选基因及耐药性检测靶标。
Objective To study the relationship of IRE-BP 1 and drug resistance of BEL-7402 hepatocellular carcinoma cell line. Methods To design primers for amplification of IRE-BP I gene fragment on the basis of known gene sequence, fluorescence quantitative PCR was used to detect the expression level difference between sensitive BEL-7402 hepatocellular carcinoma cell line and adriamycin resistant BEL-7402 hepatocellular carcinoma cell line. Results The IRE-BP 1 gene fragment was cloned successfully. FQ-PCR proved the IRE-BP 1 gene expression level of adriamycin resistant BEL-7402 hepatocellular carcinoma cell line is 3.57 times higher than that of sensitive BEL-7402 hepatocellular carcinoma cell line. Conclusion The IRE-BP 1 gene plays a certain role in drug resistance development al mechanism of hepatocellular carcinoma cell line, and it can be used as a new resistance gene and drug resistance detection target.
