摘要
目的了解湖南地区新生儿感染肺炎克雷伯菌染色体介导喹诺酮类耐药基因parC基因的流行情况。方法采用PCR法对26株肺炎克雷伯菌进行parC基因检测,采用法国生物梅里埃公司生产的VITEK2-COMPACT全自动细菌鉴定检测16种抗菌药物的体外抗菌活性,PCR扩增产物经纯化后测序并进行序列分析。结果 parC基因在所有耐药株中均有第80、91位氨基酸的改变。结论本省ICU儿童感染肺炎克雷伯菌染色体介导喹诺酮类耐药基因parC基因突变情况十分普遍,监测其突变频率、位点对指导临床用药及耐药机制研究十分重要。
Objective To investigate the prevalence of infection caused by Klebsiella pneumoniae and the mutation of chromosome-mediated quinolone resistance gene,parC gene among newborns in Hunan Province.Methods PCR method was used to detect the parC gene mutation in 26 strains of Klebsiella pneumoniae.VITEK2-COMPACT automatic bacterial identification instrument was employed to detect in vitro antibacterial activity of 16 antimicrobial agents.PCR amplification products were purified,and then identified by sequence analysis.Results parC genes in all drug-resistant strains showed amino acid changes at the positions 80 and 91.Conclusions The mutation of chromosome-mediated quinolone resistance gene,parC gene is prevalent among ICU newborns in Hunan Province.Monitoring the mutation frequency and sites of parC gene plays a very important role in guiding clinical medication and researching drug resistance mechanism.
