摘要
目的观察维生素E(Vit E)对链脲菌素(STZ)所致胰岛β细胞凋亡与Bcl-2/Bax蛋白、特异性半胱氨酸酶(Caspase)表达变化的影响。方法将分离、培养的胰岛β细胞分为正常对照组(胰岛β细胞不做任何处理)、STZ组(培养液中加入STZ)、Vit E处理组(培养液中加入Vit E 60 min后再加入STZ)。分别用电子显微镜观察细胞形态,流式细胞仪检测细胞凋亡率,免疫组化法检测Bcl-2和Bax蛋白的表达,分光光度法测定caspase-8、caspase-9、caspase-3活性。结果与正常对照组比较,STZ组细胞凋亡率及caspase-8、caspase-9、caspase-3活性均显著增加(P<0.05),Bcl-2表达下降、Bax表达增强(P<0.05)。Vit E处理组以上变化均受到明显抑制(P<0.05)。结论 Vit E能够减少STZ诱导的胰岛β细胞凋亡,其机制可能与下调Bax/Bcl-2比例、降低Caspase活性有关。
Objective To investigate the effect of vitamin E(Vit E) on apoptosis and expression changes of Bcl-2 /Bax and Caspase of Streptozotocin(STZ)-induced islet beta cells.Methods Isolated and cultured beta cells of islet were divided into the control group(beta cell of islet without any handling),STZ group(STZ was added into culture solution) and Vit E group(STZ was added into culture solution 60 minutes after Vit E addition).The cells forms were observed by electron microscope,apoptosis rate was identified by flow cytometer,the expressions of Bcl-2 and Bax protein were detected by immunohistochemical staining,and the activity of caspase-8,Caspase-9 and Caspase-3 was detected by spectrophotography.Results The activity of caspase-8,caspase-9 and caspase-3 of STZ group was significantly increased(P 0.05),the expression of Bcl-2 was reduced,and the expression of Bax was increased than those in control group(P 0.05).Conversely these values were blocked in Vit E group(P 0.05).Conclusion Vitamin E can attenuate the apoptosis of streptozotocin-induced islet beta cells,the mechanism of which may be correlated with its capability of down-regulation of Bax /Bcl-2 proportion and decreasing Caspase activity.
