IL-4、NF-κB在大鼠肝脏缺血再灌注损伤模型的表达及意义 被引量:2

摘要 目的检测白介素-4(interleukin-4,IL-4)、核因子-κB(unclear factor-kappa B,NF-κB)在SD大鼠肝脏缺血再灌注损伤模型(ischemia reperfusion injury,IRI)中的表达,探讨其与肝脏缺血再灌注损伤的关系。方法采用实时荧光定量PCR(Real-time—PCR)法定量检测缺血肝叶中IL-4、NF-κB的mRNA表达变化。健康雄性SD大鼠70只,随机分为假手术组(10只,同样行开腹手术但不夹闭血管)、缺血再灌注组(60只,建立大鼠肝缺血再灌注模型,进行45min的部分肝门阻断然后再灌注),缺血再灌注组又分为夹闭肝门45 min组,再灌注30、60、120、240、360 min组。结果在假手术组、夹闭肝门缺血45 min组及再灌注30、60、120、240、360 min组中,IL-4 mRNA的表达量分别为0.156±0.057、0.303±0.065、0.431±0.061、0.460±0.070、0.567±0.102、0.686±0.070、0.662±0.140,NF-κB mRNA的表达量分别为0.266±0.071、0.384±0.063、0.446±0.067、0.473±0.062、0.636±0.071、0.735±0.068、0.561±0.111,上述表达量随着缺血及再灌注时间的延长而升高,其中IL-4的表达在再灌注240 min后趋于平稳,NF-κB的表达在再灌注240 min时达高峰,然后开始下降。结论 IL-4和NF-κB表达量在肝脏缺血再灌注损伤模型中升高,可能与缺血再灌注损伤的发生、发展相关。 Objective To Study the expression of IL -4 and NF - κB in hepatic ischemia/reperfusion injury (IRI) model in SD rats, and discuss the relationship between hepatic ischemia reperfusion damage. Methods The liver IL -4 and NF - κB mRNA were detected by real - time quantitative PCR( Real - time - PCR) in hepatic ischemia lobe. 70 healthy male SD rats were randomly divided into sham operation group( 10 rats, performed open operation but the vessels had not been blocked) ; ischemia/reperfusion group (60 rats, establishment of liver ischemia reperfusion model in rats;the part of hepatic portal was occluded for 45min and then reperfused). Ischemia reperfusion group was subdivided into the portal clipping for 45 min group,reperfusion for 30,60,120,240 and 360 min group. Results The expression of IL -4 mRNA in sham operation group was O. 156 ±0.057 ,in occlusion of hepatic portal for 45 min group was 0. 303 ± 0. 065, in reperfusion for 30 min, 60 min, 120 min, 240 min and 360 min groups were 0.431 ± 0.061,0. 460 ± 0. 070,0. 567 ± 0. 102,0. 686 ± 0.070 and 0. 662 ± 0. 140, respectively. The expression of NF - κB mRNA in sham operation group was 0. 266 ± 0.071, in occlusion of hepatic portal for 45 min group was 0. 384 20. 063 ,in repel'fusion for 30 min,60 min, 120 min,240 rain and 360 min groups were 0. 446 ± 0. 067 ,0. 473 ± 0. 062,0. 636 ± 0. 071,0. 735 ± 0. 068,0. 561 ± 0. 111, respectively. The expression of IL -4 and NF - κB mRNA mentioned above increased with the time of ischemia and reperfusion prolonged. The expression of IL - 4 became stable after reperfusion for 240 min and the expression of NF - κB reached to the peak at 240 min after reperfusion and then began to decline. Conclusion The expression of IL - 4 and NF - κB are increased in the liver ischemia/reperfusion injury,which may be related with the occurrence and development of is- chemia reperfusion injury.
