用Infiltrator浸壁球囊导管在体转基因的实验研究

摘要 目的 评估infiltrator浸壁球囊导管在体转基因的效率、应用条件及临床实用价值。方法 用该导管将calponin基因转至兔髂动脉 ,用RT PCR法检测calponinmRNA的表达 ;通过病理检查了解在不同注射压力下进行转染基因对血管壁的损伤情况。结果  6只动物转基因全部成功 ,且血管壁未受明显损伤 ,用较大压力转基因可引起管壁局限性增生。结论 Infiltrator导管转导效率高 ,对血管壁无明显损伤 ,其应用条件基本满足了临床的要求。 To assess the efficiency,application conditions and the clinical values of gene transfection in vivo with infiltrator intramural catheter Methods Calponin was transferred into the right iliac artery walls of rabbits with the infiltrater catheter; The expressions of calponin mRNA were detected by RT-PCR method;Morphologic changes in artery wall after gene transfection under different injective pressure were observed Results Gene transfer succeeded in all 6 animals without obvious damage to artery wall Limited proliferations of intima were observed when injection pressure was higher Conclusion The gene transfer with infiltrator catheter is effetive and safe The application conditions of infiltrator have met the requirements for clinical gene delivery
