摘要 【目的】通过试验优化激素配比,获得花生根愈伤组织诱导率高的激素组合。【方法】以花生根组织为原料,采用三元二次通用旋转方法设计试验,将花生根组织接种在含不同配比的激素培养基中进行诱导,并统计其诱导率。【结果】三元二次通用旋转方法分析结果表明,回归模型R2为0.9521,所获回归模型可较好拟合愈伤组织诱导率与各参试因子之间的关系,并达到极显著水平,二次回归数学模型为:y=83.399-1.335x1-2.156x2+5.469x3-1.844x1x2-1.259x12-1.940x22-3.557x32。单因子分析结果表明,诱导率与2,4-D、KT、6-BA之间均呈抛物线关系,随着激素浓度水平的增加,诱导率表现为先增加后下降的趋势;2,4-D、KT、6-BA的最佳诱导浓度分别为2.04、1.67、3.05mg/L。2,4-D、KT和6-BA的交互作用明显,当2,4-D、KT和6-BA3种激素组合浓度分别为2.50、1.80、3.00mg/L时,花生愈伤组织诱导率达最高值,为87.55%。【结论】不同激素种类对花生根愈伤组织诱导率的影响程度为6-BA>KT>2,4-D。以MS为基本培养基,附加30g/L蔗糖、9g/L琼脂及不同浓度配比的2.50mg/L2,4-D、1.80mg/LKT、3.00mg/L6-BA,对花生根愈伤组织诱导具有明显促进效果。 [Objective]Hormone proportion was optimized to get the best combination for the callus induction of peanut root. [Method]Using quadratic rotation combinatorial design, peanut root tissue was inoculated to medium under different hormone combinations and its corresponding callus induction rate was statistically analyzed. [ Result ] The analysis showed that the regression model with R2=0.9521 could best fit the relations in callus induction rate and its effective factors and could reach the extremely significant level, and the quadratic regression model was as follows:y=83.399-1.335x1- 2.156x2+5.469x3-1.844x1x2-1.259x12-1.940xz2-3.557x32. [n single factor analysis, callus induction rate was parabolically re- lated with 2,4-D, KT and 6-BA. Along with the increase of hormone concentration, callus induction rate was increased first and then decreased; the best concentrations for 2,4-D,KT and 6-BA were 2.04,1.67 and 3.05 mg/L,respectively. 2,4-D, KT and 6-BA actively interacted with each other. When the concentration of 2,4-D, KT and 6-BA was 2.5,1.8 and 3.0 mg/L, the callus induction rate of peanut root was the highest (87.55%). [ Conclusion ]The order of the effect of three hormones on callus induction rate of peanut root was 6-BA〉KT〉2,4-D. Callus induction of peanut root could be greatly promoted cultured in MS medium added with 30 g/L sucrose, 9 g/L agar, 2.5 mg/L 2,4-D, 1.8 mg/L KT, and 3.0 mg/L 6-BA.
