摘要
为建立一种快速便捷的检测大蒜病毒的方法,首先以4个大蒜产区的蒜种为材料采用传统的RT-PCR方法分析我国大蒜的主要病毒类型,在此基础上优化RT-PCR方法,建立大蒜病毒的快速检测方法.传统RT-PCR扩增到的序列经测序分析表明我国大蒜主要病毒类型为大蒜潜隐病毒GLV、马铃薯Y型病毒和大蒜新型病毒,通过优化RT-PCR方法、扩增模板、反应体系和循环次数,结果表明采用两步法RT-PCR方法,直接以RNA酶A处理的大蒜叶片Trizol裂解液为模板,经过40-45轮扩增循环,可以扩增到我国主要3种大蒜病毒,这为大蒜病毒的快速诊断提供技术支持.
In order to establish a rapid and convenient method to detect garlic virus, the main virus types of garlic from four production areas in China were analyzed by traditional RT- PCR. A new method of detection was established by modifying traditional RT- PCR protocol. Analysis of the sequences indicated that garlic virus in China is mainly garlic latent virus, potato Y virus and garlic new virus. RT- PCR protocol, the templates for amplification, reaction system and cycle times were all modified. The result shows that three virus could all be detected by 40- 45 times amplification cycle of two- step RT- PCR protocol in which Trizol lysate of garlic leaves treated with RNase A was used as amplification templates. The novel method can be efficiently used for rapid detection of garlic viruses.
