摘要
目的:为研究Twist1和Bcl-2协同诱导上皮间充质转变(EMT)发生直接相关的microRNA(miRNA)提供线索。方法:构建Twist1和Bcl-2表达质粒,单独转染或共转染获得过表达Bcl-2、过表达Twist1和共同过表达Twist1/Bcl-2肝癌细胞模型,在此基础上结合miRNA芯片分析单独过表达或共过表达Twist1/Bcl-2可引发的miRNA表达谱变化。结果:共过表达Twistl/Bcl-2可引起肝癌细胞多个miRNA发生明显表达变化,数据结果差异具有统计学意义(P<0.01)。结论:Bcl-2与Twist1协同作用可引起广泛的miRNA发生明显的表达变化进而调控多个下游靶基因活性,从而改变肿瘤的多种生物学行为。
Objective: To provide clues for the miRNAs that directly related with the initiation of EMT caused by the cooperation of Twistl/Bcl-2. Methods:The Bcl-2 and Twistl expression plasmid were constructed. The control group, Twistl overexpressing group, Bcl-2 overexpressing group and Twistl/Bcl-2 co-overexpressing group of hepatic celluler cancer (HCC) models were established by the transfection of one or both plasmids described above. On that basis, miRNA chip was used to analyze the alteration of the miRNAs profile caused by the coexpression of Bcl-2 and Twistl. Results: The Twistl/Bel-2 co-overexpressing could obviously change the expression of multiple miRNAs in HCC, the differences among the data results had significant values (P 〈 0.01). Conclusion: Bcl-2 cooperating with Twistl can lead to obvious changes of extensive miRNAs expression, then regulate the activity of multiple downstream target genes, final- ly result in various biological behavior changes of the tumor.
