摘要
为明确东方田鼠腹腔细胞在感染日本血吸虫后的变化特征,用无菌PBS灌洗日本血吸虫感染后的东方田鼠腹腔,离心洗涤细胞沉淀,用含150mL/L小牛血清的DMEM培养基培养细胞,HE染色、瑞特染液染色和姬姆萨染色后用倒置显微镜观察细胞形态特征。与正常东方田鼠腹腔细胞相比,东方田鼠感染血吸虫后腹腔细胞种类较多,细胞形态不规则,且随培养时间的延长,有大量细胞集落出现,并依次呈现增殖、变形、崩解及纤维样的变化,纤维样腹腔细胞可传代1次。结果表明,东方田鼠感染日本血吸虫后,其腹腔细胞类型和数量发生了较大改变,为深入研究东方田鼠腹腔微环境在其抗血吸虫中的作用及进一步的体内外免疫学试验提供了技术方法和操作依据。
In order to understand the characterization of peritoneal cells from abdominal cavity of Mi- crotus fortis infected with Schistosoma japonicum, the cells of M. fortis before and after infection were iso- lated in vitro using PBS lavage method. After being washed using centrifugation, the isolated cells were cul- tivated with DMEM medium containing 150 mL/L newborn calf serum. After HE staining,Giemsa's stai- ning,and Wright^s staining, the cells were observed on morphology, nuclear-cytoplasmic ratio, and types under inverted microscope. Compared with peritoneal cell from healthy M. fortis, much more cells types were obtained from M. fortis infected with S. japonicum and their morphologies were irregular. With the extension of culture time, cell colonies appeared and sequentially presenting proliferation, deformation, dis- integration and fibroblasts and fiberlike peritoneal cells could be passaged one times only. The results sug- gested that there were differences in the type and quantity of peritoneal cells from abdominal cavity of M. fortis after being infected with S. japonicum. These results can be utilized as good references for providing technical methods and for future studies of the molecular mechanism resistant to S. japonicum infection.
