卵巢癌组织中TLR4表达及其临床意义

Expression and Clinical Significance of Toll Like Receptor 4 in Ovarian Carcinoma

目的探讨Toll样受体4(TLR4)在卵巢癌组织中的表达及其临床意义。方法采用RT-PCR和Westernblot方法,检测40例卵巢癌组织及其癌旁组织、30例正常卵巢组织中TLR4表达,并分析TLR4表达与卵巢癌临床病理特征的关系。结果 TLR4蛋白表达与肿瘤分化程度、国际妇产科协会(FIGO)分期、淋巴结转移有关(P<0.05)。West-ern blot检测结果显示,卵巢癌组织、癌旁组织和正常卵巢组织中其相对表达量分别为0.801±0.073、0.399±0.078和0.134±0.059,卵巢癌组织中TLR4表达量明显高于相应的癌旁组织及正常卵巢组织(P均<0.05)。RT-PCR检测结果显示,TLR4 mRNA表达量卵巢癌明显高于癌旁组织和正常卵巢组织,分别为0.0647±0.0253、0.0350±0.0220、0.0167±0.0136,P<0.05。结论 TLR4在卵巢癌组织中呈高表达,且其表达与卵巢癌病理分级有关,TLR4可能通过免疫机制参与卵巢癌的发生、发展过程。

Objective To investigate the expression of Toll like receptor 4（TLR4） in ovarian carcinoma and explore its clinical significance. Methods The expression of TLR4 in 40 ovarian carcinoma tissues,paracancerous tissues and 30 normal o- varian tissues were detected by RT-PCR and Western blotting analysis, respectively. The relationship of TLR4 with pathological grade,clinical stage, and metastasis of ovarian carcinoma were statistically analyzed. Results The highly expressed TLR4 was significantly correlated with the degree of carcinoma differentiation, an advanced FIGO stage and lymph node metastasis （P 〈 0.05 ）. The Western blot results showed that TLR4 protein expression in ovarian carcinoma tissues ,paracancerous tissues and nor- real ovarian tissues were 0. 801 ± 0. 073,0. 399 ± 0. 078 and 0. 134± 0. 059, respectively. The expression of TLR4 in ovarian cancer was significantly higher than that in normal tissue and paracancerous tissues. RT-PCR showed that the expression level of mRNA in ovarian carcinoma, paracancerous tissues and normal ovarian tissues were 0. 0647 ± 0.0253,0. 0350 ± 0. 0220,0.0167 ± 0.0136,P 〈 0.05. Conclusion TLR4 has a high expression in ovarian carcinoma tissues and TLR4 expression has a close re- lationship with pathological grades of ovarian carcinoma tissues. TLR4 plays an important role in the development and progression of ovarian cancer through immunologic mechanisms.
