天麻对大鼠大脑皮质神经元细胞缺氧后超微结构及Bax和Bcl-2表达的影响 被引量:7

Effects of Gastrodia elata on ultrastructure and expression of Bax and Bcl-2 within rat cerebral cortical neurons after hypoxia

摘要 目的:探讨天麻对大脑皮质神经元细胞缺氧后超微结构及Bax和Bcl-2表达的影响。方法:制备体外培养大鼠大脑皮层神经元原代培养及缺氧的模型,模型成功后随机分为正常对照组、缺氧组、天麻组(低、中、高剂量组)、尼莫地平组;采用倒置显微镜、超微病理学、免疫组化的方法,观察天麻对大脑皮质神经元细胞缺氧后超微结构及Bax和Bcl-2表达的影响。结果:①超微结构的观察:天麻可明显改善缺氧对神经细胞的损伤。②免疫组化观察:在缺氧处理前用天麻预处理可提高Bcl-2的表达,抑制Bax的表达,与缺氧组相比有显著性差异(P<0.05)。结论:天麻可明显改善缺氧后神经细胞损伤的超微结构;提高Bcl-2的表达,抑制Bax的表达,减少缺氧引起的神经细胞凋亡的发生,对神经细胞起到保护作用。 Objective: To investigate the effects of Gastrodia elata on ultrastructure and expression of Bax and Bcl2 within rat cerebral cortical neurons after hypoxia.Methods: Prepared the rats cerebral cortical neurons in primary culture and hypoxia model.The rats modeling successfully were randomly divided into control group,hypoxic group,Gastrodia elata group and Nimodipine group.Using an inverted microscope,ultrastructural pathology,immunohistochemical method,to observe the expression of Bcl-2 and Bax on cerebral cortex neuronal cells after hypoxia.Results: ①Ultrastructural observation: Gastrodia elata could significantly improve the hypoxic damage of nerve cells.②Immunohistochemical observation: The intervention with Gastrodia elata before hypoxia treatment could increase the Bcl-2 expression,inhibiting Bax expression compared with the hypoxic group there was a significant difference(P0.05).Conclusion: Gastrodia elata can significantly improve the ultrastructure of hypoxic injury neurons,Gastrodia elata can increase Bcl-2 expression,inhibition of Bax expression,reduction of hypoxia induced neuronal cell apoptosis,which has a protective effect on neurons after hypoxia.
