金欣口服液血中移行成分体外抗呼吸道合胞病毒的研究

Vitro experimental study of anti-respiratory syncytial virus effect on Jinxin Oral Liquid transitional ingredients of blood

目的:通过研究金欣口服液血中移行成分的体外抗呼吸道合胞病毒(RSV)作用,明确金欣口服液作用的物质基础。方法:采用细胞培养技术,噻唑蓝(MTT)比色法检测细胞活性,以利巴韦林为阳性对照,观察金欣口服液血中移行成分槲皮素、虎杖苷、黄芩素、汉黄芩素、异鼠李素、大黄素甲醚在人喉癌上皮细胞(Hep-2)中对RSV的抑制作用,以治疗指数(TI)为评价指标。结果:在Hep-2细胞中槲皮素、虎杖苷、黄芩素、异鼠李素、大黄素甲醚对RSV具有直接灭活作用,TI分别为12.78、38.07、28.93、9.43、1.58,槲皮素、异鼠李素对RSV感染具有一定的治疗作用,TI分别为12.00、4.25,汉黄芩素对RSV感染有预防作用,TI为1.57。结论:槲皮素、虎杖苷、黄芩素、汉黄芩素、异鼠李素、大黄素甲醚是金欣口服液抗呼吸道合胞病毒的物质基础。

Objective： To elucidate the substance basis of Jinxin Oral Liquid by analyze anti-respiratory syncytial virus（RSV） effect of the constituents in serum of rats oral adinistrated with Jinxin Oral Liquid.Methods： The effect of anti-RSV was evaluated with RSV infected human laryngeal epithelial cells（Hep-2） for transitional ingredients of quercetin,polydatin,baicalein,wogonin,isorhamnetin,physcion by cell culture technology and methyl thiazolyl tetrazolium（MTT） colorimetric assay detecting cell activity,as a positive control drug ribavirin,therapeutic index（TI） as an evaluating indicator.Results： Quercetin,Polydatin,Baicalein,Isorhamnetin,Physcion had a direct inactivation of RSV in infected Hep-2 cells,the TI was 12.78,38.07,28.93,9.43,1.58,respectively.Quercetin,Isorhamnetin had a therapeutic effect on RSV infection,the TI was 12.0,4.25,respectively;Wogonin had a preventive effect,the TI was 1.57.Conclusion： Quercetin,Polydatin,Baicalein,Wogonin,Isorhamnetin and Physcion are the material basis for anti-virus in Jinxin Oral Liquid.
