从微血管内皮细胞转运功能探讨针刺对缺血性中风的影响机制 被引量:5

Study of the effects of acupuncture from the transporting function of microcascular endothelium cell on the rats with focal cerebral ischemia

摘要 目的:通过研究针刺对脑缺血大鼠侧脑室微量注射葡萄糖转运蛋白(Glut-1)、P-糖蛋白(P-gp)阻滞剂后微血管内皮细胞转运功能的影响,探讨针刺是否通过影响微血管内皮细胞的Glut-1和P-gp的表达,减轻缺血后神经细胞的损伤,从而保护缺血脑组织,为针刺治疗脑缺血性疾病提供实验及理论依据。方法:采用大脑中动脉闭塞法制备局灶性脑缺血模型,侧脑室微量注射5-硫D-葡萄糖、环孢菌素A后,观察电针内关穴(每天1次)7d后脑组织葡萄糖、乳酸、丙酮酸、Ca2+和谷氨酸的含量;免疫组织化学方法检测Glut-1、P-gp的表达。结果:针刺可提高脑组织葡萄糖含量,降低乳酸、谷氨酸、Ca2+含量,促进Glut-1、P-gp表达。侧脑室注射Glut-1、P-gp阻滞剂后可抑制内皮细胞转运功能,减低针刺的上述效应。结论:针刺具有改善脑缺血后微血管内皮细胞转运功能,减轻缺血后神经细胞损伤的作用。针刺治疗脑缺血机制可能与脑微血管内皮细胞转运功能有关。 Objective: To study acupuncture stimulation on the transporting function of microvascular endothelial cells after the intracerebroventricular microinjection of Glut-1,P-gp blockers in rats with focal crebral ischemia.Whether acupuncture could reduce the ischemic nerve cell damage and protect the ischemic brain tissue by affecting Glut-1 and P-gp expression in the microvascular endothelial cells was explored in the present study.Methods: Cerebral ischemia animal models were induced by middle cerebral artery occlusion with thread embolus in Wistar rats.After ivc 5-thio-D-glucose and cyclosporine A,electroacupuncture was applied to bilateral ‘Neiguan’(PC 6) for seven days.The content of Glucose,Pyruvic Acid,Lactic Acid,Ca2+ and Glutamate in brain tissue was analyzed with specialized measuring kits.The expression of Glut-1 and P-gp in hippocampal CA1 area was observed with the method of immuno-histochemical.Results: The content of Lactic Acid,Ca2+ and Glutamate in brain tissue were decreased after acupuncturing at Neiguan.The content of Pyruvic Acid and Glucose was increased in Neiguan groups.Acupucture can improve the expression of Glut-1 and P-gp in hippocampal CA1 region of MCAO rats.Conelusion: The results demonstrate that EA improves the transporting function of microvascular endothelial cells and reduces the injured neurons in rats with focal cerebral is chemia.
