摘要
目的:探讨β-榄香烯对人膀胱癌5637细胞的诱导凋亡作用。方法:常规培养人膀胱癌5637细胞,分别用15.0、30.0、45.0 mmol/L的β-榄香烯作用于细胞,以未加药组为对照组,流式细胞仪检测细胞凋亡百分比的变化。此外,对40只裸鼠以皮下注射膀胱癌5637细胞进行荷瘤造模,并随机分为3个β-榄香烯处理组(低剂量组50mg/kg、中剂量组100mg/kg、高剂量组200mg/kg)和1个空白对照组(生理盐水)。每组10只,连续用药20d后,测量各组裸鼠的移植瘤的大小,免疫组织化学法及Western blot检测各组裸鼠移植瘤组织caspase-3蛋白裂解片段的变化。结果:β-榄香烯可明显诱导体外培养细胞发生凋亡,在裸鼠移植瘤模型中发现β-榄香烯高剂量处理组中荷瘤的大小明显小于空白对照组,免疫组织化学法及Western blot检测结果显示β-榄香烯高剂量处理组中caspase-3蛋白裂解片段量明显高于空白对照组。结论:β-榄香烯对膀胱癌5637细胞具有生长抑制作用,并且通过caspase-3介导的细胞凋亡途径诱导膀胱癌细胞凋亡。
Objective :To investigate the function of β -elemene to induce the apoptosis of human bladder cancer cell line 5637. Methods: Human bladder cancer cell line 5637 was treated with β - elemene in different concentration. Flow cytometry was used to analyze the rate of apoptosis. In addition, human bladder cancer cell line was injected subcutaneously into 40 nude mice which have been randomly divided into three treatment groups ( low - close group 50mg/kg,middle dose group 100mg/kg,high dose group 200mg/kg)and a blank control group (saline). The size of the transplanted tumors in nude mice were evaluated after 20 days continuous administration. The quantity of caspase -3 protein cleavage fragments were evaluated by immunohistochemistry and Weastern blot. Results: Significant dis- tance were found between the high doses of beta - elemene treatment group and the control group. Conclusion: β - elemene could induce the apoptosis of bladder cancer cell through the pathway of capase - 3.
