摘要
目的观察间充质干细胞(mesenchymal stem cells,MSCs)对SMMC-7721肝癌细胞中肿瘤干细胞的调控作用及其相关机制。方法将MSCs与肝癌细胞混合注入裸鼠体内,观察种植瘤生成情况。用MSCs条件培养上清体外培养肝癌细胞,流式细胞仪检测其CD133+细胞比例改变情况。ELISA检测MSCs条件培养上清中TGF-β的浓度。TGF-β处理肝癌细胞,流式细胞仪检测其CD133+细胞比例改变情况。结果裸鼠种植瘤实验显示MSCs在体内可以促进肝癌生成。体外用MSCs条件培养上清培养SMMC-7721肝癌细胞株可上调其中CD133+细胞比例,呈时间依赖性。MSCs分泌TGF-β至培养上清,TGF-β处理肝癌细胞可上调其中CD133+细胞的比例,并亦呈时间依赖性。结论 MSCs对肝癌干细胞具有调控作用,TGF-β参与介导MSCs的这一作用。
Objective To investigate the effect of mesenchymal stem cells (MSCs) on the regulation of cancer stem cells in liver cancer cell line SMMC-7721 and the underlying mechanism. Methods The mixture of MSCs and SMMC-7721 liver cancer cells were injected subcutaneously into nude mice. CD133 + cell content was detected by flow cytometry after cultured with MSCs conditioned medium or treatment with TGF-β. ELISA was employed to examine the concentration of TGF-β in MSCs conditioned medium. Results MSCs promoted xenograft tumor growth in vivo. CD133 + cancer cell content was increased when SMMC-7721 cells were cultured with MSCs conditioned medium or treated with TGF-β in a time-dependent manner. MSCs secreted TGF-β to the medium. Conclusions MSCs can up-regulate cancer stem cell in liver cancer cell line SMMC-7721 ,which is mediated by TGF-β.
