摘要
目的应用MR分子影像学评价壁虎活性单体BH1273209对兔VX2移植瘤的治疗作用,为BH1273209的进一步开发利用提供实验依据。方法建立兔VX2移植瘤模型,12天后筛选成瘤均匀的27只随机分成A组BH1273209高浓度组、B组BH1273209低浓度组及C组生理盐水组,14天后行MRI常规序列、EPI-DWI检查,测量肿瘤组织的DWI信号值、表观弥散系数(Apparent Diffusion Coefficient,ADC)值。MRI检查结束后处死全部荷瘤兔,将肿瘤组织固定、包埋、切片行HE染色。结果 B组肿瘤信号强度(1826.30±460.01)较C组肿瘤(1162.34±258.06)明显增高;A组肿瘤(527.99±81.98)较C组肿瘤明显减低。测量肿瘤组织的ADC值,B组肿瘤组织ADC值(0.0009±0.0001)较C组肿瘤(0.0014±0.0001)明显减低,A组肿瘤(0.0016±0.0001)较C组肿瘤明显增高。结论 MR分子成像可以准确、客观、无创的评价BH1273209对兔VX2移植瘤的作用。低浓度的BH1273209能促进兔VX2移植瘤生长,高浓度的BH1273209能抑制肿瘤生长。
Objective The effect of gecko active monomer named BH1273209 on the rabbit VX2 transplant tumors was evaluated by MR molecular imaging. Experimental basis was provided for further development and utilization of BH1273209. Methods Established of rabbit VX2 transplant tumors model, after 12 days, 27 rabbits with uniform tumor were chosen and randomly divided into three groups. Group A, BH1273209 high concentration group; group B, BH1273209 low concentration group; and group C, physiological saline group. The drug was administered via the ear vain 12 days after implantation. BH1273209 was administered once daily and the duration of the treatment lasted 14 days. MR imaging (MRI) scan including MR conventional sequence and DWI were performed. Tumor Volume and DWI signal value and ADC of the tumors were measured. After magnetic resonance imaging, the rabbits were sacrificed and the tumors were taken out. The tumor specimens were performed by hematoxylin and eosin staining. Results The tumor DWI signal intensity of group B (1826.30±460.01) was significantly higher than group C (1162.34±258.06), group A (527.99±81.98) was significantly lower than group C. The ADC value of group B tumor tissue (0. 0009±0. 0001) was significantly lower than group C (0. 0014±0. 0001), group A (0. 0016±0. 0001) was significantly higher than group C. Conclusion The effect of BH1273209 on rabbit VX2 transplant tumor was evaluated accurately, objectively, noninvasively by MR molecular imaging. BH1273209 with low concentration might promote rabbit VX2 tumor and high concentration BH1273209 might inhibit tumor growth.
