摘要
目的探讨促红细胞生成素(EPO)对兔局灶性脑缺血皮质区神经肽Y(NPY)表达的影响。方法成年健康新西兰兔42只,雌雄不限,以开颅法建立大脑中动脉闭塞脑缺血模型,随机分为脑缺血组(18只)、EPO干预组(18只)和假手术组(6只),前两组又根据缺血时间分6 h、12 h和24 h组,分别用免疫组织化学及图像分析法检测脑缺血皮质区NPY的表达改变。结果与假手术组相比,脑缺血组、EPO干预组缺血侧皮质NPY阳性细胞数量随缺血时间延长显著增多(P<0.05)。与EPO干预组比较,脑缺血6 h组NPY阳性细胞数量未见明显改变。脑缺血12 h(P<0.05)、24 h(P<0.01)组,缺血侧皮质区NPY阳性细胞数量增多更为明显。结论 EPO通过抑制局灶性脑缺血皮质区NPY的表达,可能是其实现对缺血缺氧脑组织保护作用的机制之一。
Objective To study the effect of erythropoietin(EPO) on the expression of neuropeptide Y(NPY) after focal cerebral ische- mic cortex in rabbit. Methods 42 New Zealand adult rabbits were randomly divided into ischemic group (n -- 18 ), erytbropoietin-treated group ( n = 18 ) and sham-operation group ( n = 6 ). The former 2 groups were divided into subgroups according to isehemia time after 6 h, 12 h,24 h. The expression of NPY were measured by immunohistochemistry and image analysis in each group. Results Comparing with the sham-operation group, the number of NPY positive-cells increased obviously in other 2 groups ( P 〈 0.05 ) as the ischemic time expanding. Comparing with erythropoietin-treated group, there was no significant differences in the number of NPY positives-cells of 6 h subgroup. But the number of positives cells increased in 12 h subgroup ( P 〈 0.05 ) and 24 h subgroup ( P 〈 0.01 ). Conclusion EPO has protective effect a- gainst cerebral ischemia injury in rabbits,which may be related to inhibit the expression of NPY.
