摘要
以杜梨为试材,采用同源序列法和RACE技术对杜梨SAMDC基因进行了克隆,命名为PbSAMDC,GenBank登陆号为:JX624260。生物信息学分析表明,PbSAMDC基因全长1 683 bp,编码358个氨基酸,相对分子量为39.767 7kDa,等电点为4.96;其编码氨基酸同源性分析表明,PbSAMDC基因与MdSAMDC1同源性最高,达到96%,其次是葡萄、甜橙等,同时对该基因的生物活性等进行了讨论,研究对梨属植物SAMDC基因的逆境调控机制和抗逆基因工程育种具有重要意义。
In this study,S-adenosylmethionine decarboxylase,designated as PbSAMDC(GenBank accession number:JX624260),was isolated from Pyrus betulaefolia Bge.by homologous cloning and RACE technology.The results showed that the full-length cDNA sequence of PbSAMDC is 1 683 bp and encodes 358 amino acids,molecular weight and isoelectric point of the protein is 39.767 7 kDa and 4.96 respectively.And the deduced amino acids homology analysis indicated that PbSAMDC has a high homology with MdSAMDC1(96%),and then the grape,sweet orange.In the same time,biological activity of PbSAMDC was discussed in the end.The research has an important theoretical and practical significance for developing stress regulation mechanism of SAMDC and stress-tolerance breeding of pear.
