Necdin-Wnt及STRA6-JAK2-STAT5信号通路与肝星状细胞活化的关系研究

摘要 肝纤维化的发生与多种信号通路密切相关,有研究证实Wnt信号通路促进肝星状细胞(hepaticstellate cell,HSC)活化。Necdin作为经典Wnt信号通路的上游信号分子,其与Wnt10b启动子结合,激活Wnt信号通路,通过抑制PPARγ,促进HSC活化。STRA6-JAK2-STAT5信号通路可以通过正向调节PPARγ维持HSC的静止。因此,研究Necdin-Wnt和STRA6-JAK2-STAT5信号通路与HSC活化之间的关系,有助于为肝纤维化临床治疗提供一些新的思考。 Hepatic fibrosis has a close relationship with multiple signaling pathways. Previous studies have established that there is a link between HSC activation and Wnt signaling pathway. Necdin, the upstream molecule of canonical Wnt signaling pathway, binds the Wntl0b promoter in activated HSCs via the inhibition of PPAR7. The STRA6-JAK2-STAT5 signaling pathway may maintain HSC in a static state via up-regulation of PPARy. This review focuses on the action mechanism of Necdin-Wnt as well as the relationship between STRA6-JAK2-STAT5 signaling pathway and HSC activation.
