1株角蛋白酶产生菌的筛选、鉴定与产酶特性研究 被引量:3

摘要 采用平板透明圈筛选法,从长期堆积羽毛废弃物的土壤及沼渣中筛选出22株产角蛋白酶的菌株,其中B1-2菌株的酶活较高,经16S rRNA鉴定为红灰链霉菌(Streptomyces rubrogriseus)。对B1-2菌株的发酵工艺进行了优化,试验结果表明,B1-2菌株产角蛋白酶的最佳条件为:发酵时间96 h,发酵温度35℃,接种量8%;最佳发酵培养基为:5%羽毛粉角蛋白,0.1%磷酸氢二钾,0.01%MgCl2,0.5%蛋白胨,1.5%玉米粉;最佳试验条件下酶活性达到132.4±2.48 U/mL。
