摘要
目的探讨20-羟基蜕皮甾酮(20E)对糖尿病心肌病的保护作用。方法 Wistar大鼠60只随机分为正常组、模型组、对照组和治疗组,每组15只,后3组大鼠用60mg/kg腹腔注射链脲佐菌素建糖尿病模型,之后分别灌服蒸馏水、5mg/kg盐酸二甲双胍和10mg/kg 20E溶液,药物干预6周后,观察心脏形态学改变,测定空腹血糖、心脏糖基化终末产物(AGE)、糖化血红蛋白(HbAlc)、HDL-C和LDL-C含量,实时荧光定量分析线粒体酶complexⅠ、Ⅱ、Ⅲ和Ⅳ的表达。结果与模型组比较,治疗组心肌损伤程度减轻,空腹血糖、AGE、HbAlc和LDL-C分别降低了62.4%(P<0.01)、38.8%(P<0.05)、51.2%(P<0.01)和47.9%(P<0.01),complexⅠ、Ⅲ和Ⅳ表达水平分别增加了2.51倍(P<0.01)、2.81倍(P<0.01)和2.08倍(P<0.01)。结论 20E对糖尿病心肌病具有保护作用。
Objective To study the role of hydroxyecdysone(20E) in protecting rats against diabetic cardiomyopathy. Methods Sixty Wistar rats were randomly divided into normal group,model group,control group and treatment group(15 in each group). A diabetes mellitus model was established by injecting streptozocin into abdominal cavity at the dose of 60 mg/kg. Six weeks after the rats in model group,control group and treatment group were given distilled water,metformin (5 mg/kg) and 20E(10 mg/kg) respectively,their heart morphology was observed and their set um levels of glucose(Glu), advanced glycosylation end (AGE) products, HbAlc, HDL-C and LDL-C were measured. Expressions of complexes I -IV were detected by RT-PCR. Results The myocardial inury was milder in treatment group than in model group. The serum level of Glu, AGE products,HbAlc and LDL-C was 62.4%, 38.8%, 51. 2% and 47. 9% lower, respectively, after treatment(P〈0.05,P〈0.01). The expression level of complexes Ⅰ , Ⅲ ,Ⅳwas 2.51-fold,2.81- fold and 2.08-fold higher,respectively,after treatment(P〈0.01). Conclusion 20E can protect rats against diabetic cardiomyopathy.
