摘要 筛选不同表型的胃癌细胞亚株。方法:用体外粘附法筛选对层粘素(laminin,Lin)有不同粘附力的MKN-45胃癌细胞亚株,并用Boyden小室法和裸鼠接种法观察细胞亚株的体内、外侵袭转移能力。结果:经体外反复以层粘素粘附筛选,得到高粘附力的MKN-45Lm+细胞亚株和低粘附力的MKN-45Lm-细胞亚株。Lm+细胞体外对人工基膜Matrigel的侵袭、移行能力及在裸鼠体内的成瘤性和肺转移灶形成率均大于Lm-细胞,并有高转移和高侵袭倾向。结论:体外粘附法能成功筛选特定表型的胃癌细胞亚株,胃癌细胞的粘附力与侵袭转移相关。 Background/Aims: To screen the subpopulations of gastric cancer cell line with different malignant phenotype. Methods: Two subpopulations from human gastric cancer cell line MKN-45, i.e.less invasive, non-metastatic; more invasive and metastatic subgroups were separated by laminin (Lm) adhesion method in vitro. The invasiveness and migratory capacities between the two subgroups were observed by Boyden chamber and by inoculation into nude mice. Results: The two subgroups, laminin adherent cel1s (Lm+) and laminin nonadherent cells (Lm ) were separated. The invasiveness and migra tory abilities in vitro were higher in Lm+ cells than those in Lm- cells. The rate of tumor formation by subcutaneous inoculation and the rate of lung tumor foci formation by tail-vein inoculation were higher in Lm+ cells than those in Lm cells. In vivo, Lm+ cells were found to have higher metastatic potential and higher invasiveness. Conclusions: In vitro, the adhesion method offers an advantage of simple, time saving and high successful rate in screening the particular phenotypic cell subpopulation.There are discrepancies in invasiveness and migratory abilities in vitro between Lm+ and Lm- cells,which suggest such behaviors of gastric cancer cells are closely related to their adhesiveness to base-ment membrane and extracelluIar matrix.
