摘要
目的 :研究咪唑安定 (midazolam ,MZ)对急性分离大鼠海马CA1区锥体神经元γ 氨基丁酸受体A型(GABAA)的作用。方法 :采用制霉菌素穿孔膜片钳技术。结果 :无GABA时 ,MZ不能单独诱发内向电流 ;GABA能诱发可被GABAA 受体拮抗剂荷包牡丹碱 (bicuculline,BIC)阻断的电流IGABA,0 0 1~ 30 μmol/L能增强IGABA,使GABA浓度效应曲线平行左移 ,但不改变IGABA的最大反应值 ,用标准细胞外液洗脱后 ,电流可恢复至给药前水平。结论 :MZ不能直接激活GABAA 受体 ,但能通过增强GABA的作用调控该受体 ;海马CA1区的GABAA
Objective:To investigate the effect of midazolam(MZ) on the γ aminobutyric acidA(GABA A) receptors in acutely dissociated rat hippocampus CA1 pyramidal neurons.Methods:Using nystatin perforated patch clamp recording configuration.Results:In the absence of GABA,MA alone could not induce inward currents.GABA induced inward currents(I GABA ) could be blocked by GABA A receptor antagonist,bicuculline(BIC).MZ could argument I GABA over the range of 0 01 30μmol/L and cause the concentration response curve for GABA to the left without affecting the maximum response of I GABA .Conclusion:MZ can not directly activate GABA A receptors,but can positively modulate the receptors by augumenting GABA response.GABA A receptors in the hippocampus are probably principal targets of MZ.
