摘要
观察蛇毒复合酶对人肝癌细胞 BEL - 74 0 2体外生长的抑制作用。经体外细胞培养 ,采用台盼兰染法、细胞形态观察和流式细胞术 ,检测人肝癌细胞的生长抑制率 ,以及细胞形态和细胞周期的变化。结果 ,蛇毒复合酶对人肝癌细胞有极强的抑瘤作用 ,8h和 12 0 h抑瘤率分别达 79.5%和 94 .5%。镜检及涂片染色发现癌细胞胞膜破裂、胞质外溢。流式细胞术分析显示蛇毒复合酶主要杀伤 G0 / G1期人肝癌细胞 ,同时 ,阻滞 S期细胞进入 G2 / M期。说明 :蛇毒复合酶对人肝癌细胞具有一定体外抑瘤作用 ,其作用机理与破坏细胞胞膜和干扰细胞生长周期有关。
Objective:To investigate the inhibition of venin complex enzyme on human hepatocellular carcinoma BEL-7402 in vitro.Methods:The rate of inhibition and the alteration of cell form as well as cell cycle were analyzed by trypan-blue tropochrome assay,morphologic observation and flow cytometry in vitro.Results:The venin complex enzyme exhibited significant antitumor activity on BEL-7402.After 8 and 120 hours,the rates of the inhibition were 79.5% and 94.5% respectively.The ruptered cytomembranes and cytoplasm flux were found by microscopy and smear cytologic examination.With analyzed by flow cytometry,the venin complex enzyme mostly killed the G0/G1 stagcells,and blocked the S stage cells developing to be G2/M stage cells.Conclusion:The venin complex enzyme had a definitive antitumor effect on BEL-7402,of which mechanism were correlative with the destruction of the cytomembranes and the interference in cell growth cycle.
