摘要
目的 :在大量筛选实验的基础上 ,选择一种能短时间内判断温缺血断肢再植时限的方法。方法 :应用沸重蒸水沸法提取、生物发光技术连续测定兔离断肢体骨骼肌 ATP的含量 ,同时对每一标本作病理检查。结果 :温缺血条件下兔离断肢体骨骼肌 ATP含量下降程度与骨骼肌的变性程度呈对应关系 ,当 ATP含量下降 95%以后 ,骨骼肌发生不可逆变性。结论 :应用生物发光法可在短时间内 (约 0 .5h)判断温缺血断肢的再植时限 ,是一种有前途的检测方法。

Objective:To explore a new method that can evaluate the critical ischemic time of the amputated limb in a relatively short time.Methods:The muscular tissue of the amputated limbs of 10 rabbits was homogenized in boiling double-distilled water followed by boiling,abstraction and assayed by the bioluminescent method.The muscular ultrastructural cell degeneration of each biopsy was under light-microscopic and scanning electron microscopic analysis at the same time.Results:The amount of ATP in the muscular tissue directly correlated with the degree of muscular cell degeneration.When 95% ATP was lost,the muscle got an irreversible necrosis under which circumstance the limb could not be replanted. Conclusion:The bioluminescent method can be used in deciding the critical time of the amputated limb in a short time (about 0.5 h) and the method is worth studying further. 〔
